Boosting osimertinib Blood Brain Barrier penetration in patients with epidermal growth factor receptor mutated non-small cell lung cancer (OSIBBBOOST)
Description
Osimertinib is one of the preferred first line treatment options for metastatic epidermal growth factor receptor (EGFR)-mutated non-small cell lung cancer (NSCLC). Whereas central nervous system penetration of osimertinib should be sufficient to reach therapeutic concentrations, drug transporters such as breast cancer resistant protein (BCRP, also known as ABCG2) may affect intracranial concentrations.
In addition to this, patients with NSCLC and brain metastases at baseline who received osimertinib in combination with platinum-doublet chemotherapy had better progression-free survival as compared to those treated with osimertinib monotherapy. This raises the question whether intracranial osimertinib exposure may be influenced by addition of chemotherapy.
This proof of concept study aims to evaluate the effect of febuxostat (an ABCG2 inhibitor) or platinum-doublet chemotherapy in combination with osimertinib on intracranial osimertinib exposure, which will be measured as osimertinib concentrations in cerebrospinal fluid.
