A single arm phase II study evaluating intracranial efficacy of Sacituzumab govitecan (SG) with bevacizumab in patients with asymptomatic brain metastases from non-small cell lung cancer (NSCLC) (SG and beva brain)

Description

About 10-20% of patients with non-small cell lung cancer (NSCLC) present with brain metastases at baseline, and the incidence of brain metastases increases to about 40% over the course of the disease. Unfortunately, no good systemic treatment options exist for patients with progressive intracranial disease after chemotherapy in combination with an immune checkpoint inhibitor.

Therefore, this trial combines sacituzumab govitecan (SG) plus bevacizumab in patients with brain metastases from NSCLC. SG is an antibody-drug-conjugate targeting TROP2, which is overexpressed in lung cancer among other types of cancer. Bevacizumab inhibits angiogenesis by binding vascular endothelial growth factor (VEGF) and thereby preventing the activation of VEGF receptor (VEGFR). 

This single-arm phase 2 study aims to evaluate the intracranial efficacy of this combination of SG and bevacizumab in patients with non-squamous NSCLC and untreated or progressive asymptomatic brain metastases.
